@NikoMcCarty In Australia, this is one of the few bacteria we can make GMOs in (eg add a plasmid, make a KO) where it is exempt from writing a dealing to do the work. Unfortunately, it is mislabelled as K12 in legislation, which is a capsule type… see Part 2: legislation.gov.au/F2001B00162/la…

TIL that E. coli K12 — the most widely used strain of bacterium for biological research — is named "K-12" because it was first isolated from a diphtheria patient staying in room K-12 of a Stanford University hospital in 1922. Naming conventions in biology are weird!

Do y'all do this too when you wanna save tubes but still run a small sample of your PCR reactions on a gel before purifying? 2.5uL Purple Loading Dye 10uL PCR Sample Pipette up and down in puddle and load immediately.

@etowah0 Where do I start? Forgot: 1) To check amplicon oresent 2) PCR clean up 3) restriction digestion, 4) ligation, SLiCE, or gibson assembly steps 5) much better to do chemical transformation instead of electroporation when cloning. Otherwise, forgot to dialyse before electroporation
