Here, we discuss the potential for the development of analgesics, structure-activity relationships on NaV channels, biosynthetic hypotheses and chemical syntheses.

It is a great pleasure to share that our extensive review on "The Chemistry and Biology of the Tetrodotoxin Natural Product Family" has been accepted for publication at Angewandte Chemie: doi.org/10.1002/anie.2….

But that is not all! To showcase the potential of dfdc azobenzenes for the development of chemical tools, we synthesized dfdc-OptoBI-1 and demonstrated its biological activity as a red light-operated activator of TRPC6 channels in HEK293 cells.

The data was used to establish a computational approach that allows to compute realistic UV-vis spectra by combining TD-DFT excited-state calculations with MD simulations. Our combined study enables the in silico design of new highly red-shifted photoswitches.

Very excited to see our work on photoswitchable menthol (AzoM) to control the cold receptor TRPM8 published! Our chemical probe allowed the analysis of current kinetics, and the roles of individual amino acids in channel gating, AzoM binding and modulation by PIP2.
