This cover image shows a MERFISH-based subcellular map of a human sebaceous gland, visualizing the spatial distribution of transcripts from 278 distinct genes that delineate the stages of sebocyte differentiation. Each dot represents an individual RNA molecule captured at subcellular resolution, revealing the spatial segregation of basal, transitional, and lipid-producing sebocytes within the gland lobules. The tissue was processed using tangential sectioning to maximize the number of intact sebaceous glands, followed by gel embedding, enzymatic digestion, and extended clearing to optimize signal detection for MERFISH imaging. The MERFISH experiment was performed by Vizgen, and the resulting data were visualized using the proprietary MERSCOPE Visualizer software (.vzg files). This approach enabled precise mapping of transcript distributions within the gland, providing an orthogonal validation of the sebocyte differentiation stages identified in our scRNA-seq and Stereo-seq analyses.
