Revealing the epigenetic modulator lysine-specific histone demethylase 1a as a new target for kidney diseases doi.org/10.1172/jci.in… Here, Tobias B. Huber @Tobias_B_Huber & team @UKEHamburg use a state-of-the-art, tour de force experimental design to show LSD1 regulates kidney development, and its dysfunction disrupts key kidney cells, leading to cyst formation in mouse and organoid models. The image shows severe structural changes in the adult mouse kidney with loss of KDM1a. #Nephrology #KidneyDisease

The increased thermogenesis in the formerly white adipocytes was NOT from UCP1. Rather, up to 80% of thermogenesis was from the futile creatine cycling @kazaklab . Credit to #Kelly_Long and #Cheyl_Cero.

Here the authors propose that treatment of adipocytes with BMPs results in an energy expending phenotype without transdifferentiation into beige adipocytes, providing a new approach to treat #obesity related metabolic disease. @JCI_insight insight.jci.org/articles/view/…

Is p53 mutational status alone enough to predict poor prognosis among patients with #AML? doi.org/10.1172/jci.in… Here, ASCI member Sami N. Malek & team @UMichMedSchool quantitatively define patients with AML who have WT TP53 but defective TP53-mediated apoptosis, revealing that this group has a dismal long-term survival. The figure shows survival of patients with AML stratified by biomarkers.

A new model for alveolar macrophage-Mycobacterium #tuberculosis (#TB) interactions! doi.org/10.1172/jci.in… Michael S. Glickman @MichaelGlickma1 & team @MSKCancerCenter establish a stem cell–derived lung alveolar macrophage model to reveal itaconate functions differently between human macrophage types in the host defense against Mtb: The figure: Brightfield and stained images of two macrophage populations.

Long QT syndrome is a life-threatening #cardiac #arrhythmia for which current therapies don’t correct the molecular defect doi.org/10.1172/jci.in… Alfred L. George, Jr. @nupharm1 & team @NUFeinbergMed now identify the first trafficking corrector for KCNQ1 mutations that cause #LQTS—the small molecule VU0494372 increases KCNQ1 cell surface levels, total levels, and surface trafficking efficiency in a dose-dependent manner in mammalian cells. The images show VU0494372 increases cell surface and total KCNQ1.

This Research Letter offers a creative approach to #sepsis based on a naturally occurring efferocytosis-enhancing RNA doi.org/10.1172/jci.in… Small noncoding RNA TY2 enhances efferocytosis and improves outcomes in a mouse model of sepsis by @AleCiulloPhD, @Marbanlab, @TheIbrahimlab & team @CedarsSinai. The image shows exposure to TY2 increases E. coli clearance by bone-marrow derived macrophages as indicated by fluorophore as a reporter of efferocytosis (green).

In #ALS, bone changes have largely been attributed to disuse. doi.org/10.1172/jci.in… Francesco Roselli @RoselliLab & team now report accelerated senescence of bone cells before the onset of neurological symptoms, due to reduced sensitivity to osteogenic mediators, in a mouse model of ALS. The figure shows distal femurs with weakly mineralized bone (red), moderately mineralized bone (yellow), and highly mineralized bone (blue) in WT and ALS model mice (SOD1) at presymptomatic (p45) and symptomatic (p110) stages, in which early trabecular thinning and reduced mineralization precede cortical bone loss.
