Supramolecular chemistry of collarenes—Collarenes are belt-shaped hydrocarbons composed of fused benzenoid and 1,4-cyclohexadiene rings, offering a fascinating platform for supramolecular chemistry. In a new study reported in @J_A_C_S, Zhang, Guo, Huang et al. demonstrate multimodal complexation between [20]collarene and a range of fullerenes, leveraging the host’s rigid yet adaptable cavity. The resulting host–guest complexes exhibit promising ultrafast charge separation and a remarkably stable charge-separated state. Full article: pubs.acs.org/doi/10.1021/ja…

Crystallizing the uncrystallizable—PROTACs are new drug molecules in the beyond Rule of Five (bRo5) chemical space. They suffer from extremely poor aqueous solubility and low crystallizability due to their unique structure. This complexity makes it particularly challenging to analyze PROTACs from a solid-state preformulation perspective. Now, Screen, Steed @jonathan_steed , et al. report in @J_A_C_S a comprehensive crystallization protocol used to grow single crystals of a cereblon-recruiting PROTAC called “AZ1.” Their work resulted in the determination of two structures: an anhydrous form and a nonstoichiometric p-xylene solvate, achieved through 3D electron diffraction and synchrotron X-ray crystallography. This study paves the way for solid form screening and a deeper understanding of pharmaceutical systems that operate in the challenging bRo5 space. Full article: pubs.acs.org/doi/10.1021/ja…
