@JoshTycko Thus, I would expect that the longer blast gene effectively silences the gRNA via a wake of positive supercoiling that inhibits transcription of the gRNA...want to hear more? Come to mSBW!

CRISPR vector design Q: Hill et al. says this design with convergent Pol2 and Pol3 transcripts has 3x reduced editing efficiency; they hypothesize Blast transcription inhibits sgRNA expression (Nat Meth 2018). Anyone know of other data from convergent designs? Thx! 🧬⭕️✂️

@Leili_Rohani Are the cells all coming from one source, eg PSC-derived, or do the cells you want to stop proliferating get added in separately? For the former you could consider something like a cell classifier (@LauraProchazka). They can be tricky but powerful for things like this.
