T cell engagers for I&I will have a lot of clinical readouts this year. This update (report from MS) from $AMGN on blinatumomab last week doesn't bode well for the class and it's unclear if next gen formats will have an advantage beyond dosing regimen. $CGEM will have an important clinical update in RA and SLE this Q.

Late line CRC therapies in development following $CTMX's data this week (from Guggenheim). A lot is riding on $CTMX's 10 mg/kg cohort which had just 19 evaluable patients (3 patients were excluded so ITT response rate is lower) so this needs to be corroborated in the dose optimization stage. Temab-A's ORR is probably in the 15-20% range in the broad CRC population (data in the table are for MET amplified CRC), making $CTMX's ORR superior for now.

Remarkable patient anecdotes - a wonderful scientific achievement $REGN Bummer that these programs faded into the obscurity of large pharma, where they will undoubtedly garner less coverage and visibility than if $DBTX and $AKUS were still standalone companies "A year after treatment in one ear with DB-OTO, a child born profoundly deaf was able to enjoy music, engage in imaginative play and participate in bedtime reading when the cochlear implant on their other ear was removed. These seemingly small interactions are life-changing for these children as well as their families"

Updated data from $REGN on its DB-OTO gene therapy for otoferlin-related hearing loss: investor.regeneron.com/news-releases/… 10/11 children w/ least 1 assessment post treatment experienced improved hearing; some dramatically so. Story to come later.

Did the market expect $DYN to radically outperform $RNA? DM1 drug candidates look pretty similar and suddenly Dyne is heading into registrational cohort with 32 pts on 6M endpoint vs Avidity's 150 ph3 trial on 30w endpt. Time to market looks very close.

@ohadhammer And yet Otezla still doing > 2bn/yr. What happens when efficacy is cranked up drastically with manageable safety? IMO, these markets are so large that there will always be room for some uptake of orals, while perhaps not largest market share.

Great comparison from Morgan Stanley's analysts of new and next gen therapies for psoriasis. I think oral drugs (esp Tyk2 but also "oral biologics" like IL23 and IL17) will have a hard time competing with approved and half-life extended IL23 antibodies. Small molecules are unlikely to obtain the selectivity and safety profile seen with antibodies imo...

Pretty wild the divergence of fortunes in $GILD and $VRTX over the last decade Close to a decade ago Incivek was discontinued, and VRTX was forced to focus on their CF franchise Since then, VRTX has over quadrupled in stock price, GILD is down like 37%, lower mc than VRTX

@BertrandBio @JacobPlieth The idea of bispecific ADCs continues to intrigue and could be really exciting. On the Hexabody tech, would imagine it likely that toxin conjugation would interfere with complement recruitment and internalization. If HexElect could be ADC-ified, that would be huge.

$GMAB swinging for the fences this morning. Great to see European big biotech dip its toes in meaningful M&A. $ 1.8bn sure is a significant ticket for a phase I/II FRa ADC, one of the most crowded targets around.
