non-specificity for disease, but specificity regarding mechanism? - pTau elevation in blood as a cellular stress response to the deposition of amyloid

A positive blood tests for #alzheimersdisease might indicate systemic amyloidosis instead, suggesting clinicians should be cautious when interpreting results. @uni_tue ow.ly/xJ5h50YtUGk

Elevated serum pTau levels are not specific to Alzheimer´s disease and may also serve as diagnostic tool for the most common types of systemic amyloidosis, with potential utility in distinguishing amyloidosis-related polyneuropathy from polyneuropathy of other etiologies @MathiasJucker nature.com/articles/s4159…
English

@EricTopol yes, non-specificity for disease, but perhaps specificity regarding mechanism: pTau elevation as a cellular response to the deposition of (any) amyloid
English

The blood neuromarkers p-tau181 and p-tau217 that can predict and diagnose Alzheimer's disease are also abnormal in certain types of amyloidosis, indicating both some non-specificity and potential help in diagnosis of these conditions

@louisanicola_ @EricTopol ...this overlap could actually be informative, because abnormal p-tau in these patients may reflect shared amyloid related pathophysiology affecting neuronal integrity...very true!!
English

It highlights an important limitation of p-tau181 and p-tau217 as Alzheimer’s biomarkers, since tau phosphorylation can also increase with systemic protein misfolding and neuronal stress seen in amyloidosis. At the same time, this overlap could actually be informative, because abnormal p-tau in these patients may reflect shared amyloid related pathophysiology affecting neuronal integrity.

it is out! Elevated blood p-tau 181 and 217 levels are not specific to Alzheimer disease and may also serve as a diagnostic tool of ATTR and AL amyloidosis! #alzheimer #biomarker. Wonderful collaboration with 3 European amyloid centers.

Blood #neurofilament (NfL) could serve as a cross-species biomarker for #aging interventions and #mortality prediction | PLOS Biology plos.io/46iNxiX. -- To better understand mortality predition across an even wider range of species, we are now looking for more samples!

Neurofilament Light (NfL) may serve as a cross-species blood biomarker for assessing ageing interventions and predicting mortality. It was great fun to measure NfL in all these speceis, even in a crocodile!

We discovered these astrocytic inclusion >25 y ago (DOI: 10.1126/science.1542796); Very satisfying to finally see the genetic linkage. Great work. The Smarcal1-Usp37 locus modulates glycogen aggregation in astrocytes of the aged hippocampus: Cell Systems cell.com/cell-systems/f…
