@jptosar We think that gymmosis is largely driven by increased protein binding of oligos containing phosphothioates. Modified oligos such as nusinersen happily enter the cell but struggle to escape the endosomes. Here increased nuclease stability might allow them to form a leaky depot

3) Gymnosis is not an ASO-specific mechanism... It's how cells sense naked or nonvesicular exRNAs. Modified bases or LNPs do, in addition to other tricks, what we did with RI here: protect exRNAs from degradation!
