We are excited to announce the release of @manager_micro v2.0.2_20230815, which includes a new feature in the “On-The-Fly Processor Pipeline” menu: Deskew! Broadly, this is useful for automatically deskewing, rotating, and projecting light sheet images in Micro-Manager. (1/7)

We present long-term stabilization of an oblique plane microscope (OPM). By stabilizing the remote focus unit of an OPM, we can enable long-term acquisition without light-sheet drift. The feedback signal comes from a laser, no fluorescence is expended. biorxiv.org/content/10.110…

@Nano_Juli @AndriyChmyrov @AndrewGYork @amsikking has been tackling thermal stability a lot recently. He's posted some of his data, which suggests that the monolithic block could be a good approach. I think he's planning to try out monolithic blocks soon, so stay tuned! x.com/amsikking/stat…

Thermal expansion -> a typical problem in time-lapse microscopy at high resolution. Here I measure the axial shift of the focal plane of a Nikon 40x0.95 air objective at ~1.6um/K, or about ~1.5x the depth of field per degree C: github.com/amsikking/Niko…

At the heart of every snoutscope is a sub-micron alignment between two objectives. 1°C thermal drift kicks the lightsheet out of focus and destroys your image ☹️ Our optics room swings by 5°C/day 😭...so @AndrewGYork & I made a crossbraced O2/O3 subunit to passively compensate!

PSA: Heat-set inserts make stronger threads in a 3D print, and look quite tidy.

@aandr314 Huh weird. I think that's just a reflection but I'll check it tomorrow. Only thing it's seen is a < 5 mW laser

Following a poke from @RunningPhoton, I've fixed a bug in my KL divergence calculation function which means our parameter-free deconvolution approach now *really* has no hand-tuned parameters at all: beryl.mrc-lmb.cam.ac.uk/rlgc_notebook/ Feedback always welcome. x.com/JamesDManton/s…

We (@AndrewGYork, @RunningPhoton, @microRussell and I) are still working on the manuscript describing our automatic parameter-free deconvolution approach, but we've made a Google Colab notebook where you can try it out without needing to install anything: beryl.mrc-lmb.cam.ac.uk/rlgc_notebook/

Made a prototype light trap out of aluminum shim stock and Musou black paint. Watching paint dry has never been so interesting. The matte finish is fantastic!

@richardfuisz Yep. You should use an airbrush for best results, but it's still pretty black even with a paintbrush.
