Biggie lacked depth. Pac made gangsters cry with words. Two different levels of art.

This is a continuation of the war/military motif. The live spot he's referring to is the Don/general position. It’s “live” because it’s an active, powerful, and recognized leadership position of authority that already carries influence and respect. It’s a position of greatness that comes with great responsibility. Only certain types of individuals are built to handle it. This is a prime example of the military mind he used to say he had. He’s saying that although he has the top spot, he continues to build on what he already has and doesn’t become complacent, which is an easy trap to fall into in his position. This is the mentality the best generals and Dons have, and that’s how you keep your spot.
Connection to the previous line:
In the line before this, he said that his Mob controlled all cash from Atlantic-Pacific, showing that they are very powerful. The money they control plays a huge role in the power he and his mob have. The money and power he has gives him access to more resources, and can also make him more respected and feared. With that amount of cash and power, he’s able to strengthen his military and strike fear into people, making him hard to kill. He tapped into this "military mindset" even more so after being set up, robbed, and shot. Surviving that definitely had something to do with why he viewed himself as being hard to kill.

He’s also referring to the top spot in the rap game, which he held. The spot is live because it’s a powerful position of greatness, a position that’s tough to uphold because there’s no two-man spot; only one can reign at the very top and be number 1. You can’t literally build with a live spot; it’s figurative. Rap is a very competitive sport, and the competition is always gunning for your spot and trying to knock you off your throne. You must stay on your Ps and Qs and continuously build on what you've already built. Makaveli did this, and it kept him laser sharp. This prepared him for war and made him hard to kill lyrically.
This is also directed at Live Squad; he felt like Stretch was disloyal, disingenuous, and was involved in setting him up at Quad Studios where he was robbed and shot five times. This is why he asked GOD (Father) and his father (Billy Garland), “How the hell did I survive these five shots?” in the very next line. He used an allusion to create this wordplay.
He juxtaposed the words “live” and “kill” to create an antithesis. The words are related, but they’re polar opposites. Live means anything that possesses life; alive, living, as opposed to “dead,” and kill is obviously something you do to unalive something or someone that’s alive. The juxtaposition of “live” and “kill” was all by design.
When he said, “We claim Mob, M.O.B., if you'll be specific, we control all cash from Atlantic-Pacific,” it’s a play off the acronym “Money Over Bitches.” He juxtaposed the word "cash" with this acronym to create amplification, a rhetorical device, which was used to help form the wordplay.

“Kill and build” form an monosyllabic internal rhyme, while “live spot” and “five shots” form an external multi. This is a prime example of rhyme sandwiching.
