Every approved orexin drug on the market blocks OX2R. OX2R-004 activates it. The entire class of approved orexin drugs, suvorexant, lemborexant, and daridorexant, are small-molecule antagonists designed to block OX2R for insomnia. No approved orexin agonist exists anywhere and clinical-stage candidates in development are small molecules for narcolepsy only. @BioProtocol's scientific team and @peptai_ ran OX2R-004 through the full 9-gate autonomous discovery pipeline on BIOS infrastructure in just 24 hours. The candidate is an 18-residue peptide agonist designed as a selective OX2R activator that entered genuinely uncharted therapeutic territory, targeting a receptor that every existing drug approaches in the opposite direction. Computational results across all eight gates show: • Molecular weight: 1,980 Da • Strongly hydrophilic profile • Zero strong immunogenicity signals • Toxicity score: 0.0 Two remaining Gate 8 risks have known peptide engineering solutions. Every gate decision, tool call, pass, and failure is published openly on Molecule Labs. The pre-IND computational feasibility assessment is published on Molecule mainnet, making it the first autonomous AI drug candidate in this class with on-chain provenance at pre-IND stage. Wet-lab validation costs under $1,500, while traditional pharma spends millions and years to reach the same checkpoint The next step is a BLI binding panel at Gingko, commissioned and funded autonomously via x402. The first on-chain wet-lab attestation via @Molecule_sci Labs is targeted for Q2 2026, the first time an autonomous agent commissions, funds, and verifies a real experiment fully on-chain. This is what autonomous drug discovery looks like when it is actually running.
