@PhilipAhern @d_bayik Honestly we throw it all in at once, i.e. we have Fc block in our staining cocktail because 1) it’s higher affinity and 2) we save 30 mins, we throw the live dead in there too, and it works great. And personally I think our staining looks just fine 👍🏻

Another flow question. I was taught to Fc block cells, remove Fc block, & stain. But I see lots of people keep the Fc block in during the staining too. The latter makes sense (staining ab. could displace Fc), but is there a reason the former is advisable? Does it actually matter?

This one is definitely a ‘Phage’ turner! @harrison_oj lab used phage display screening to identify antigen-specific BCRs in mLN and PPs & used tetramer stain to track these cells! A much-needed advance in the field to fish specific B cell clones and study them in depth #Immunity
