Many, many thanks to all my co-authors, the incomparable @AnnaMolofskyLab, and the kind, thoughtful manuscript reviewers who made this work better. Questions about the data welcomed here, or via email!

This was a tough project to snag a phenotype, but it was also satisfying to do a mechanistic dive into a hit from a transcriptomic screen.

Check out the paper for all the details, including a rewarding collab w/ Mercedes Paredes doing a comparative analysis of NE development in mouse vs. human brain.

Finally, we also found behavioral deficits in sensorimotor integration (prepulse inhibition, balance beam) tasks in adult β1 cKO mice, though again this was seen only in female mice.

In a fun in vitro collab with Marlesa Godoy (Ye Zhang’s lab), we observed that stim of α1 or β receptors make astros branchier, while stim of α2 receptors make astros less branchy (this effect is *fast*, happening w/in just 3h!).

We then moved in vivo, and made an early postnatal astro-specific β1 KO mouse model. Cortical (but not white matter) cKO astros were smaller and expressed more GFAP. Surprisingly, this phenotype was specific to female mice.

We found that both mouse and human astros express all subtypes (α1, α2, & β) of adrenergic receptors, with some interesting region- and age-specific differences.

For the aficionados: beautiful previous work from @AnnaMolofskyLab (DOI: 10.1126/science.aal3589) showed transcriptomic diffs btwn developing gray vs. white matter astros. Here, we followed up on one of those diffs: higher expression of adrenergic receptors in gray matter astros.

The punchline: we found a new signaling cue - β1 adrenergic signaling - that promotes gray matter astrocyte development in mice.

While on my OBGYN rotation, I've inexplicably found myself humming "the baby's coming" part from youtube.com/watch?v=TkZyRo… rather a lot :) (but don't worry-no green babies... yet!)

Hey #zebrafish friends! I'd like to do an experiment where I image the same FOV in an agarose-mounted fish pre- and post- drug treatment (keeping fish on the scope whole time). Is this possible? Have you done this? Will chemicals penetrate agarose? cc @damiandn @jraslab

This is such a *wonderful* resource for image analysis! Loving the embedded questions to build out your intuitive understanding of the concepts
Pete Bankhead@petebankhead

If you want to learn about #bioimageanalysis I've written a free & open textbook that tries to help: bioimagebook.github.io Thanks to the wonder of @ExecutableBooks & other modern magic it's not quite like a normal book... (thread) @OpenEdEdinburgh @NEUBIAS_COST @BioimagingNA

@SchmooBlake Am not a serious runner, so curious to hear if others have thoughts, but I agree that running is v hard on the body!! My main form of cardio is biking (stationary + outside), which I find more tolerable & equal endorphins
