@Prokaryota Most heat shock ecoli will take up a purified vector if you vortex for 10s and plate immediately. It is inefficient for transformation but efficient time. You only need one colony to expand.

What's your favorite quick-and-dirty lab technique that still works for you? I make E. coli electrocompetent cells straight from plate streakouts (no pre-culture in liquid) 😱 at room temperature 😱 with just water (no glycerol) 😱. Success rate >90%, don't @ me
