I am a first time founder working on my second fundraise I’m spending the next 2 weeks setting up a lot of intro calls with seed stage investors. I’m genuinely confused about one thing: What exactly counts as “competitive” to your portfolio? Because in early-stage deep tech, stuff is confusing: - Same sector, completely different architectures - Same AI approach, completely different applications - Same modality, one builds tools, the other runs an internal pipeline - Nothing in common on paper, but both end up going after melanoma The category overlap can be zero and the market overlap total. Or the category overlap can be total and the market overlap zero. what are you actually pattern-matching on? what to watch out for here? I’d love to hear from investors of any kind.

An SF based VC told me: "Seed in SF now means anything below $50M valuation"🤯 Btw, Nikhil's point is correct - the lowest cap in the current YC W26 batch is $30M and median dilution is ~10% (i.e. $3M to $4M rounds) And, this isn't a YC specific thing - non YC co's in SF as well are raising Seed rounds at $30M to $50M In my 5 yrs since starting investing, I'd say "History may not repeat but it sure does rhyme" "Pre-Seed has become Seed" (by early 2020s; $15M val; led by solo GPs) "Seed has become Series A" (now; $40M val; led by platform funds & supersize Seed funds)

the default yc round this batch (W26) seems like 4m on 40m I remember when I first started in venture exactly three years ago (W23 batch) and most venture ppl were complaining about YC pushing their founders to do 2m on 20m in 3 years the market went from a very begrudging 2 on 20 to a more neutral 4 on 40 interesting to think about where things land 3 years from here

i just spotted a blind spot that's costing startups millions in churn. we exploited it - and hit 70% customer retention. early on, we noticed something in our user data: our users weren't monolingual. they spoke english at work. spanish with friends. hindi with parents. portuguese with their partner. and most voice ai tools forced them to pick one. so we decided to build differently. today, wispr flow supports 100+ languages. because we saw what happened when we tested other languages with our early users: they had the biggest smiles on their faces. most products treat non-english languages as second-tier experiences. but we've always believed in letting people express themselves in their own voice. if you grew up speaking tamil at home, that's how you think. that's how you express yourself most naturally. so having to switch to english just to use a productivity tool? that's not just inconvenient. it's making you feel like the way you naturally communicate isn't valid. so we built support for 100+ languages early on. the feedback we get from users in non-english languages is different than any other feature. it's not "this is useful." it's "thank you for seeing me." that's when we knew we made the right call. when you build with empathy, you don't just translate words. you meet people where they are. and that's how you build something people don't just use - they love.

The famous SFFA case treated Indians and East Asians as a single group. This masked significant heterogeneity: It's way harder to get in if you're Indian! In Columbia's internal admissions database (h/t @cremieuxrecueil), East Asian applicants had a 41% lower odds of admission than equally qualified White applicants, whereas South Asian applicants had 63% lower odds.

Pancreatic cancer ranks among the most lethal cancers, with tumors often developing rapid resistance to therapies, contributing to dismal long-term outcomes. In a major preclinical advance, scientists at Spain’s National Cancer Research Centre (CNIO), led by Mariano Barbacid, have shown that a triple-drug combination can completely eliminate pancreatic tumors in mouse models while preventing resistance from emerging. The treatment targets the KRAS signaling pathway—mutated in about 90% of pancreatic ductal adenocarcinomas—at three separate points, effectively blocking the cancer's ability to adapt and survive. The regimen combines daraxonrasib (an experimental KRAS inhibitor, also known as RMC-6236), afatinib (an approved EGFR/HER2 inhibitor used in certain lung cancers), and SD36 (a selective STAT3 protein degrader). This multi-pronged attack shuts down key downstream, upstream, and parallel pathways that KRAS relies on for tumor growth and evasion of single-agent therapies. Tested across multiple orthotopic mouse models of pancreatic ductal adenocarcinoma (including those with KRAS/TP53 mutations), the combination induced complete and durable tumor regression, with no detectable cancer recurrence even months after treatment ended and no major toxicities observed. In some cases, tumors vanished entirely, leaving no trace. While single KRAS inhibitors show promise, resistance typically develops quickly as tumors activate bypass mechanisms. By hitting three independent nodes simultaneously, this strategy overcomes that hurdle in animals, offering a blueprint for more effective targeted therapies. Pancreatic cancer's five-year survival rate remains under 10%, underscoring the urgency of such innovations. Although human trials are not yet feasible and further optimization is needed to translate these results to patients, the study represents a landmark step toward making one of the deadliest cancers potentially controllable.
