SOHM's ABBIE™ Platform: Engineering the Next Generation of Non-Viral CAR T-Cell Manufacturing SOHM's ABBIE™ platform is being developed as a non-viral approach to CAR T-cell engineering, designed to enable site-specific genomic integration without inducing double-strand DNA breaks. By fusing integrase with catalytically inactive Cas9 (dCas9), the platform is engineered to combine the permanent gene integration capability of retroviral integrases with the programmable targeting of guide RNAs. Our current development program is focused on optimizing the core integration mechanism, including linker architecture, cofactor interactions, and construct geometry, to achieve enhanced on-target performance. In parallel, we are building out the genomic characterization and functional assay infrastructure needed to rigorously validate targeting specificity, integration efficiency, and CAR T-cell cytotoxicity as the platform advances. ABBIE represents a platform-level opportunity, with multiple configurations under development for potential licensing across therapeutic applications.
