My “kitchen cabinet” isn’t a group of DC advisors. It’s a a group of Hartford middle schoolers that live in my neighborhood and I meet with regularly. We met today and talked about the federal budget. They were surprised we spend more on the military than their education!

Much earlier than expected, it is time for another short thread on a recent preprint at bioRxiv (Yang et al., doi.org/10.1101/2025.1…) by a group of authors led by Dr Anjali Kusumbe. Today, I want to cover some of the bioinformatics results presented in their preprint. As a reminder, based on the reanalysis of our scRNA-seq data from Koh et al. (PMID: 39537918), it is claimed in Fig. 1 and the accompanying text that lymphangiogenic markers are downregulated in aged skull samples. We now had the chance to closely examine the relevant data, which has uncovered a couple of major issues. 1/4

@ralfhadams @jonykipnis Our experience has been the same. It’s very, very difficult to sort these cells—and contamination is abundant—if not done carefully. I am hoping for better yields next month, adhering to protocols derived from Jony’s lab🤞 Appreciate this thread—something useful again on X!

Why is it that on my way to Japan on @AmericanAir I pay $35 for internet, which is down 80% of the time and the remaining 20% it is extremely slow, but on my way from Japan on @JAL_Official_jp, I pay $18, and the internet is solid and fast the entire flight? And I am not comparing the food and the service, because there the scale is totally different...

I'm not sure - like anyone else, I'm only listening to the news and talking to people I meet and at NIH, but ~40% of people I talk to think 44% cut will happen, ~20% think no cut will happen and politicians will fix it (I doubt it) +40% think something in between (~20% cut to NIH for the NIH Fiscal Year starting Oct 1 2025). The plan I saw from NINDS (link below) assumes the cut will go ahead, see page 5, so now (after my conversation on Friday with the NIA PO) it looks to me that there is some evidence that it is being built into current decisions re funding between now and Sep 30 - officeofbudget.od.nih.gov/pdfs/FY26/inst… as well as the known multiplier effect of a cut on new grants [1] which depends on the planned ratio of new to continuing grants which I simulated here [2] and is also in the NINDS FY26 plan above [1] x.com/PTenigma/statu… [2] x.com/PTenigma/statu…

I spoke with an NIH program officer yesterday who said that, given the massive 44% cut planned for NIH, they were only planning to award about 12 new grants before the end of the year, which are mainly ones in the 1st and 2nd percentile, so the 3 grants we had with good scores (usually fundable: 4th percentile, 13th percentile, and Impact Score=21) are not likely to be funded. 🤯The logic was that they are trying to maintain funding for clinical trials that are already in progress, as well as funding for ADRCs (30 or so Alzheimer's Centers across the U.S.). This also means that noncompeting continuations (of active grants) are likely to be awarded to the extent possible (which would be great if we had any), i.e. keep those going rather than award new grants. 🤯One group of people replying to the thread below (about limiting grant submissions) think that PIs who write a lot of grants are gaming the system, but most of the grants we get funded are on about the 3rd attempt (3rd submission) and we used to win about 1/4 of them after 3 tries, so it would take 12 tries to finally win one R01, often after many years of improving it. And in some of the R01s where I was PI, I would allocate 90k-100k USD to my own lab, while helping several other labs to get money (which is presumably a good idea, as our studies are collaborative). 🤯The new rule might actually be a good idea (I can't tell, several +'s and -'s are noted in the thread below) but it's not like we're winning them very often...

The NIH has decided that scientists can only submit 6 grants a year. OK. But you need a score that’s < 10% at least to get funded. Meaning, at best, 1 out of 10 grants you submit will have a chance (YMMV of course). 🫠

Physicians are often late and they always have a great excuse of “I had a patient”. Surgeons, and especially neurosurgeons, think it’s mauvais ton being on time… Those of us with PhDs need to come up with an excuse equivalent to “I had a patient”/“OR took much longer”… Ideas?
