How can we make PARP inhibitors more effective in castration-resistant prostate cancer? @FrancescaBZNY et al. identify the DNA repair gene LIG1 as a sensitizer to PARP inhibitors in multiple tumor models: jci.org/articles/view/… @FraLo05 @GiuliaFracassi_ @CIBIO_UniTrento @AIRC_it @PCF_Science @ERC_Research

💡Additional Insights: * LIG1 loss to be considered/analyzed for/in PARPi treatment * Additional markers to be studied, including EME1 loss and FAAP24 loss

🔑 Key Findings: * LIG1 loss is synthetically lethal with PARP inhibition. * LIG1 is altered in 6% of CRPC patients. * Combined LIG1 and PARP inhibition triggers replication stress and DNA damage, ultimately leading to apoptosis. * LIG1-PARP synthetic lethality is conserved across various cancers, including prostate, lung, breast, and colorectal. * Co-targeting LIG1 and PARP selectively impacts PCa and breast cancer cell viability.
