A study long in the making! Initiated at WashU and completed at Miami, our work demonstrates that the functional role of ion channel proteins can differ significantly depending on the type of neuron in which they are expressed.

TSC1 LOF mutations are monogenic causes of ASD. Mice with Purkinje neuron-specific Tsc1 deletion have several ASD-like behaviors. Here, Sam and Achintya (co 1st-authors) show these PNs have problems with AP firing due to a loss of Na+ channels at the AIS pubmed.ncbi.nlm.nih.gov/40015654/

At @SFN23, Sam Brown presented his dynamic clamp experiments showing the resurgent sodium current (INaR) does not affect the firing rates of central neurons. Instead, INaR tunes the availability of persistent sodium current. Check out the preprint: biorxiv.org/content/10.110…
