Check out our preprint on the Doa10 E3 ligase, the key player in ERAD-C. We combined cryo-EM, MD simulations (another cool collaboration with JC Gumbart and Diane Lynch!), and biochemical approaches to reveal how Doa10 recognizes its substrate proteins. biorxiv.org/cgi/content/sh…

Our data suggest that the large circular, enclosed transmembrane domain architecture of Doa10/MARCHF6 is to prevent non-substrate proteins from accidentally being ubiquitinated.

Updates: This is now published in Nature Communications. nature.com/articles/s4146… In revision, we tested a few more substrates, including TA proteins. All seems to interact directly with the "lateral tunnel" we identified.
