I am so excited to share our new paper in @Nature: the first programmable, site-specific integration of a large DNA payload into T cells in vivo. A single IV injection results in therapeutic levels of TRAC-targeted CAR T cells in multiple models. #Ack1" target="_blank" rel="nofollow noopener">nature.com/articles/s4158… a 🧵

Today we report single-cell APEX-seq (scAPEX-seq) — a new method for unbiased mapping of *subcellular* transcriptomes at single-cell resolution. This approach reveals cell states that are not detectable by standard scRNA-seq, and enabled us to identify regulators of CAR T function that improve solid tumor killing. biorxiv.org/cgi/content/sh…

An honor to host the one and only @EricTopol. Of course, we talk about medical futurism. And, of course, we talk about Super Agers (his latest book). But we also talk about his journey from medical student to world’s best known cardiologist to medicine’s foremost soothsayer!

@TLeavery @LocasaleLab Same here. But electricity, trash, custodial services, hazardous waste disposal are part of research. And govt has contracting standards that require unis be able to do expensive things like dispose of hazardous waste, track dangerous chemicals, and track financial compliance.

Universities with large academic medical centers are not small charities struggling to keep the lights on. They are part of sprawling financial complexes involving hospital systems, real estate empires, entertainment franchises, venture funds, and multi-billion-dollar endowments that often operate like hedge funds. Annual revenues at many of these institutions can approach or exceed $10 billion. Yet when the research funding model is questioned, the discussion suddenly turns to who will pay for janitors and light bulbs. Meanwhile, new research institutions can be run far more efficiently — potentially with overhead closer to ~15–25% of direct research costs.
SJ Williams@sjwill99

@LocasaleLab Who pays for the lights, repairs, janitorial, capital improvements, repairs and other running costs?

Excited to share our new @Nature paper! We developed an IL-2–TGFβ co-agonist that enables efficient induction of antigen-specific pTregs and establishment of immune tolerance in vivo. An important step toward programmable immune tolerance. rdcu.be/e72lN

Finally out - fully-defined, feeder-free production of CD4+ helper T cells from induced pluripotent stem cells (iPSCs)! Turns out you need to turn Notch down.... a notch 🥁 And you need to fine-tune stimulation of the T cell receptor ⚖️ cell.com/cell-stem-cell…

Tumor immune evasion is programmable🛡️🧠. Our integrated single-cell CRISPR screening framework maps kinase control of T cell–driven tumor states in Glioblastoma(GBM) 🧬and nominates druggable nodes 💊 (e.g., EPHA2/PDGFRA) that blunt evasive programs and boost T cell killing 🔥.

@ScienceKyle Okay, then it may be reasonable. I have not read the paper since it is behind a paywall. Still, it seems like an unusual target, given that it is fairly common among immune cells.

There is an obvious contradiction in this model: it claims CD70 is largely absent from normal adult tissues, yet develops a more sensitive anti-CD70 CAR T cell to detect levels below conventional detection limits. #science #immunology science.org/doi/10.1126/sc…
