@MadS100tist @kaufman_lab @chrmosimann We’ve continued putting a polyA signal in the 3’ position mostly out of habit and not totally knowing what is most effective in terms of recruiting polyadenylation machinery (e.g. if there are distance constraints). So all of our 3’ clones include the polyA.

#Zebrafish friends - Love Tol2 Kit. Having trouble lately with ? SV40 elements in some cloning (using Gibson, less Gateway these days). Suspect repetitive sequences to blame. Any good alts for 302/SV40 for 3' segment of typical prom:CDS:polyA (394/395 already have polyA)?
