The 2025 Nobel Prize in Physiology or Medicine has been awarded to Mary E. Brunkow, Fred Ramsdell, and Shimon Sakaguchi for discovering how the immune system prevents itself from attacking the body — a process called peripheral immune tolerance. Our immune system defends us from germs, but sometimes it can mistakenly target our own cells, leading to autoimmune diseases. Sakaguchi discovered special “peacekeeper” cells, called regulatory T cells (Tregs), that act like brakes to stop this self-attack. Later, Brunkow and Ramsdell found that a gene called FOXP3 controls these Tregs — and when this gene doesn’t work, the immune system turns against the body. Their groundbreaking work revealed how the body maintains inner balance and opened the door to new treatments for autoimmune diseases, cancer, and organ transplants. 🧬🏅 #NobelPrize #Science #Immunity

TBXT is present in many cancers, especially chordoma, but barely expressed in normal tissue. Aiming to exploit this difference, Cassian Yee and colleagues at @MDAndersonNews have found that TBXT peptides are expressed on MHC receptors and are now developing cell therapies to target this cancer-associated TBXT peptide-MHC complex. The first of these therapies will soon be tested in a clinical trial led by @APConley77 through MDACCs new sarcoma SPORE. Huge thanks and congrats to all involved for this exciting milestone for chordoma! Thanks in particular to @GorlickRichard for leadership of the SPORE. We look forward to seeing how the trial unfolds and continuing to work together to bring TBXT targeting immunotherapies to fruition.
