I passed my thesis defense yesterday, “Understanding #SARSCoV2 antigenic drift using complete genotype-to-phenotype maps.” It has been an absolute privilege to be a graduate student in the @jbloom_lab at the @fredhutch and @UW_MSTP.

Awesome to see more high-throughput antibody escape tech come on board, particularly from industry - where resulting data can more quickly be used to evaluate, engineer, and pool mAbs under clinical development. Great job @AAlphaBio !

Which therapeutic antibodies retain binding against emerging CoV-2 variants? Our latest pre-print is out! 33 monoclonal antibodies (including 9 in clinical trials) tested for binding against 1000's of CoV-2 variants. biorxiv.org/content/10.110… [thread 1/4]

We've created an interactive website to visualize >100,000 experimental measurements of how mutations to #SARSCoV2 RBD affect binding by antibodies & sera: jbloomlab.github.io/SARS2_RBD_Ab_e… Explore it to examine a wealth of information about the antigenic effects of viral mutations. (1/n)

I've long advocated the use of DMS-approaches to understand escape from monoclonal and pooled antibodies during their development as antiviral therapeutics, as well as during viral surveillance to guide their clinical use. Here's a great example why! science.sciencemag.org/content/early/…

Lastly, these data on BG505 Env trimer responses - and parallel unbiased mapping approaches - are of keen interest given these immunogens are currently being evaluated in multiple clinical trials. I'm excited to see what the binding and neutralizing responses in humans may be!

Our new work with @WardLab1 & PJ Klasse comparing the binding and neutralizing antibody responses to HIV Env elicited by BG505 SOSIP trimer immunization in rabbits is now available, and summarized here:

In a new study (biorxiv.org/content/10.110…), we have completely mapped mutations to #SARSCoV2 that escape human antibodies, and shown that these "escape maps" predict how virus evolves under antibody pressure & inform design of escape-resistant antibody cocktails. (1/12)
