Congratulations to Dr. Adrienne Boire, a neuro-oncologist at Memorial Sloan Kettering Cancer Center (MSK), and Dr. @TuomasTammela, a cancer biologist at MSK, on receiving @AACR Trailblazer Cancer Research Grants, announced during #AACR26. Nine early-stage investigators and six mid-career investigators were awarded grants of $1 million each to support meritorious and promising cancer research. This morning, MSK physician-scientist shared research during the Opening Plenary Session at #AACR26 about how cancer cells escape targeted therapy by changing identity. @AACR Key conclusions: ➡️ Adenocarcinomas with RB1 loss are at risk ("primed") to undergo NE lineage transition in response to therapelic stress. ➡️ In "primed" tumors, extracellular matrix production by cancer associated fibroblasts restrains NE lineage transition through integrin -> YAP/TEAD activation. Notch pathway activation acts as a second level of restraint. ➡️ This adeno <-> NEPC transition can be toggled in either direction using existing pharmacologic tools, with potential therapeutic implications. ➡️ New vulnerabilities arise once the NE transition is complete, notably on MYC family paralogs with co-option of TIP60/KAT5 as a coactivator.
