@cureffi @ThanosDim3 Dauting indeed. Yes we did. All credit to @ThanosDim3 The struggle was real. Also lots of support and discussions with internal safety committee. That was one of the reasons for using splitseq, it was potentially compatible with bsl3 environment. Happy to share specifics

@EmmanuelleVire @ThanosDim3 Wow, cool. Did you set up the entire single cell pipeline in the prion lab? We do single cell sequencing of non-prion samples in our lab but when we considered doing it on prion samples we found the infrastructure requirements rather daunting.

@tjm_sci helped visualizing these findings using spatial technology. And finally we performed RNAseq in frontal cortex from sCJD patients and healthy controls and found overlapping signatures between mice and human diseases.

29 resurgent genes show an oscillating pattern of dysregulation whereby affected at 20dpi and then again at 120dpi and onwards. Early genes are only affected at 20dpi. Late genes showed dysregulation at 120dpi and onwards.
