Thank you, Steven... we are very excited about the results because they show that, in real time, surgeons can use approaches like this to determine where and when tumors are being resected, and they might have "missed" a hidden metastasis just outside the margin of removal. The "margin" is the safe distance into which one must cut to statistically feel confident that all the tumor is removed. In complex cancers like head and neck cancer, the tissue is so complex (tongue, veins, muscle) that it is surgically challenging not to damage nearby structures. Techniques like those Professor Guolan Lu and our colleagues developed at Stanford enable more precise surgery as well as later analysis of where a therapeutic antibody can access, or what the cancer is doing to erect barriers that block access to therapeutic drugs. This helps us figure out what we need to do to break down such barriers.

6/8 💡The SSP platform is broadly applicable to antibody-based therapeutics, including immune checkpoint inhibitors, antibody-drug conjugates, and multi-specific antibodies.  Incorporating SSP into early-phase clinical trials could advance the development of next-generation cancer therapeutics by uncovering mechanisms of response and resistance, guiding individualized dosing and identifying spatial biomarkers for participant stratification.

5/8 💡 These results suggest that improving antibody therapy may require not only better targets, but also strategies to overcome spatial barriers within the tumor microenvironment.
