PSA: Stop calling macrophages in tissue M1 or M2. These are not states that exist in biology. The only use of M1 is a macrophage cultured ex vivo with LPS and IFNg; and for M2: IL-4/13/10. Macrophages in tissues are highly complex and diverse and do not resemble either of the aforementioned M1/M2 states. M1/M2 language causes confusion and sets the field back. Refer to your macrophages by the molecules they express and the cytokines they make.

It sucks to have NIH grants that arent funded. A huge waste of time. At the same time, I think cutting DEI grants is a needed correction. Much of this work is descriptive, and, at this point redundant, the few interventional studies are often non randomized, or have poor control arms, limited power, or measure proxy rather than clinically meaningful endpoints. Since the topic is feel-good among liberals, the scientific standards are often lower. It's sad to waste time writing the grant but that is a broader failure of NIH. Millions of human hours have been lost from NIH hurdles. Perhaps more hours are spent writing grants than doing science among elite investigators. The system has to change. It needs a dramatic reform. In the process, inevitably, some researchers will be disappointed. That is unfortunate, I agree.

The first NIH grant I've ever written, with my dream mentor and dream topic, just disappeared. Eliminated today, just like that, thanks to the recent 'Executive Order on Ending Radical and Wasteful Government DEI Programs and Preferencing' Hundreds of hours of work, just gone.

@jbhuang0604 I guess I don’t consider image models the same as image-text models. There are a number of instances where self supervised models work fine on images alone. This of course depends on what you want to accomplish.

Why is self-supervision in vision still not working? 🤔 When pretraining a transformer on TEXT-only data by predicting the next tokens, we see clear improvement trends as we scale the model, data, and computing. But after trying to pretrain a transformer on IMAGES-only data with contrastive learning (e.g., SimCLR), masked prediction (e.g., MAE), and self-distillation (e.g., Dino), at the end of the day, the most successful vision encoders still have to rely on vision-language paired data (e.g., CLIP. SigLIP). Or have people not yet tried connecting MAE/Dino with LLMs? Why is that? Thoughts?

To those who work with primary monocytes or PBMCs, do you have any idea what these coin-shaped features might be that are scattered amongst the immune cells? Are they platelets? They are calcein AM positive (viable) and are present at seeding time. #PBMC #Macrophage #immunecells
