Folate can be paradoxically deficient even when serum folate levels (reflecting methylfolate) are high.
This is caused by many nutrients being deficient — vitamin B2, B3, B5, B6, B12, vitamin C/E, serine, iodine, molybdenum, phosphorus, selenium, iron, sodium, copper, magnesium, zinc, and more — either directly or indirectly.
This leads to a phenomenom known as 'methyl trapping,' where methylfolate is stuck in the folate cycle and can't move forward into the MTRR step where it donates the methyl group to cobalamin, which then further transfers it to homocysteine, forming methionine, allowing for the methylation cycle to work in synergism with the folate cycle, like 2 cogs turning correctly in a well-oiled machine.
NADPH (thus its cofactors in synthesis; magnesium, vitamin B3, zinc, ATP/phosphorus, vitamin B1, and glucose) acts as the electron donor to reactivate cobalamin/vitamin B12.
FMN and FAD (thus its cofactors in synthesis; thyroid hormone/T3 so tyrosine/phenylalanine, iodine, selenium, zinc, glutathione, iron, and copper, alongside molybdenum, magnesium, and ATP/phosphorus) are anchored to the MTRR enzyme and facilitate the electron transfer to the cobalamin center.
Anything that affects thyroid hormone function (eg., cortisol, pro-inflammatory cytokines, circadian disruption) may disrupt methylation by lowering Mg—ATP and riboflavin's activation into its active forms (mentioned above).
Vitamin B12 requires optimal parietal cell function for stomach acid and intrinsic factor release for its absorption, alongside haptocorrin from the salivary glands (thus its cofactors; being choline, chloride, vitamin B5, zinc, vitamin A, potassium, bicarbonate, and anything related to healthy nervous system function).
A low redox/high inflammatory state also plays a role in methyl trapping, as this puts downward pressure on NADPH and FMN availability, oxidizing B12/cobalamin faster than MTRR can recycle it, whilst shifting homocysteine towards cysteine synthesis to make glutathione, or taurine and sulfate secondarily. So the substrates for glutathione (cysteine, glycine, glutamine/glutamate), choline/betaine, vitamin C, vitamin E, selenium, polyphenol-rich foods, CoQ10, and other redox-supportive compounds are key.
Infection and inflammation where WBCs (especially neutrophils) are on high demand for proliferation may also deplete B9/B12. Same applies for RBC turnover.
Creatine, choline/phosphatidylcholine, betaine/TMG, taurine, glutathione/NAC, melatonin, and avoidance of excessive B3 supplementation (especially niacinamide or nicotinic acid) can help spare the methylation load.
Glycine can be hit-or-miss when it comes to supporting methylation and context is key. Whilst it's also important to consider genetical defects and adjust nutrient dosages accordingly (eg., PEMT, MTRR, MTHFR, CBS).

Well s boulardii for example reduces leaky gut, increases DAO and is anti-fungal and anti parasitic.
Improving these things transiently, can allow environment to adapt, or for you to make other moves to improves the environment.

Many of your beneficial bacteria are “beneficial” for this exact reason.
Low beneficial bacteria = leaky gut.
The paradox is that with leaky gut, we have a compromised garden bed for beneficial bacteria.
