@Ines_Marin_G @globalimmuno We are using MRL/lpr lupus model. It's a genetically triggered model. Similar findings regarding TLRs in other models, as I reviewed in the intro. The subject of the work and the talk is to figure out what's different between the TLRs. I showed that TIR domains are part of it.

@globalimmuno @MShlomchik 2- which model are you using? what is triggering lupus in it? Is it known WHY one of the TLRs is protective and the other pathologic? Only the intracellular signaling? Different recognition of different nucleotides (from… self? Infections?) may have a role? THANSKS!!!

@Ines_Marin_G @globalimmuno I think that B cells driven by TLR7-dependent autoAg (e.g. RNA, like you suggest) are presenting Ag and activating autoreactive pathogenic T cells and secreting inflammatory cytokines

@Ines_Marin_G @globalimmuno Turns out you don't need autoantibodies at all for glomerulopathy. Chan, et al. 1999. A novel mouse with B cells but lacking serum antibody reveals an antibody-independent role for B cells in murine lupus. J Exp Med 189:1639-1648.

@Dr_Shuli @globalimmuno For sure these mutants are functioning in other cells and we have not deconvoluted that, but it should be studied, perhaps collaboratively with others that specialize in those cells.
