Significant weight reduction across all tested doses over an 80-week period Eli Lilly's TRIUMPH-1, a Phase 3 clinical trial evaluating retatrutide, an investigational triple hormone receptor agonist (GIP, GLP-1, and glucagon) for adults with obesity or overweight. Key Weight Loss Outcomes The trial demonstrated significant weight reduction across all tested doses (4 mg, 9 mg, and 12 mg) over an 80-week period: 12 mg Dose: Participants lost an average of 70.3 lbs (28.3%). Notably, 45.3% of these participants achieved a weight reduction of 30% or more. 9 mg Dose: Participants saw an average weight loss of 64.4 lbs (25.9%). 4 mg Dose: Even at the lowest dose with a single escalation step, participants lost an average of 47.2 lbs (19.0%). This dose also saw a lower rate of discontinuation due to adverse events compared to placebo. 104-Week Extension: For participants with a baseline BMI ≥35, weight loss continued through 104 weeks, reaching an average of 85.0 lbs (30.3%) for those on the 12 mg dose. Metabolic and Health Impact Beyond weight loss, retatrutide showed significant efficacy in moving participants out of the obesity category and improving overall health markers: Obesity Threshold: By week 80, 65.3% of participants on the 12 mg dose reached a BMI of <30, effectively falling below the threshold for obesity. This included 37.5% of participants who started the trial with class 3 obesity (BMI ≥40). Cardiovascular Improvements: The drug led to significant improvements in several risk factors, including waist circumference, non-HDL cholesterol, triglycerides, systolic blood pressure, and high-sensitivity C-reactive protein (hsCRP).
