In this paper we show compatibility with cysteine and n-term profiling. However, this workflow should be compatible for profiling any residue that has an established activity-based probe (and there's a lot of them). 6/

This reagent isn't just for show either. In a head-to-head comparison with traditional cysteine profiling using TMT we show decreased variance with sCIP-TMT likely due to not having to carry each sample through an individual sample prep. Also saves a lot of pipetting time! 5/

The cool thing about this reagent is it's future proofed and applicable to all commercially available, NHS-based isobaric tags. So whether you use TMT, TMTpro, or iTRAQ this one reagent can be utilized. 4/

To achieve this we developed a single reagent that could be in-situ conjugated to TMT reagents converting them to enrichable, click-compatible, isobaric tags. Notably, we use these conjugates directly for click without purification. 3/

It's no secret TMT labeling is a bit laborious and we saw an opportunity to streamline this by developing click-compatible isobaric tags which allow for early-stage sample combination (doi.org/10.1021/jacs.3…). However we wanted to make this technology more widely applicable 2/
