Out in @PNASNews now, we find that the Rag-Ragulator heptamer anchors the GATOR supercomplex to the lysosome. Our results highlight the complex role the Rags play in regulating mTORC1 activation and lysosomal homeostasis (1/2)

@ATinyGreenCell I actually work on metal-biology in my PhD so I have quite some experience making hundreds of slightly different trace metal containing M9 media... 😅 Precipitation can indeed be a pain (I learned that the hard way). I guess this is why they use EDTA here.

@QuastJP Some of the stock solutions are temperamental at high concentration to make 1000x or 100x. The iron ones specifically are a pain. You can make a trace mineral solution but that is borderline witchcraft and needs like a week of aeration. Note the highlighted word here, lol.

I hate volumetric measurements. A "tablespoon" of fertilizer per gallon is a jank-ass number. Gonna round it to 3g/L and start from there... That's Jack's Classic 20-20-20 general purpose fertilizer. Gonna do some comparisons for my static hydroponics. Dilutions a plenty!

@ATinyGreenCell Makes sense! Why do you mean it is hard to make the stocks of the trace elements?

@QuastJP Yes definitely! Several labs have their homebrewed list but basically it's somewhere between M9 and MS media. The nutrients mostly track oceanic trace mineral levels + nitrates and usual suspects (Mg, Ca, P, S, etc). The trace elements are important but hard to make stocks of.

@ATinyGreenCell I am curious on what you grow them. Is this agar? Any specific temperature or humidity?

Falcoponics Testing out reusing 50mL tubes as standing hydroponic systems using rockwool and a 3D printed collar. Tobacco, brassica, and mimulus to test. I left space for a tubing line for automated watering too. Fun times ahead!

@QuastJP I've tried it in my Shiny app for qPCR analysis and seems impressive. Thank you. BTW, it fully supports facet_wrap().

@QuastJP Fantastic! Much needed. Does it support interactivity? A mouse click on one plate to get more details would be a terrific add on.

I am actually not surprised by this since we always see that rampamycin is very specific based on validation experiments for our LiP-MS method. We have tons of data on this. Would you be interested in having a look at that to maybe include it? Just DM me!
