@RobynBarbato I normally grow the culture to stationary phase (O/N for E. coli) then dilute 1:1,000 fold into fresh media to get very low OD culture. That way, you get the full growth phases of lag, log, and stationary.

Fantastic colleagues, I need some help over here! For growth curves of bacteria, do you pre-incubate in liquid culture? If so, which growth phase is best before inoculating cultures for the growth curve analysis - logarithmic or stationary? Why? Please include helpful references

@RobynBarbato Anecdotal, but from my PhD work (which involved many many growth curves with hot spring bacteria to test for Hg resistance), the growth curves were only reproducible when started from a log phase culture.

Intriguing happenings @ the #permafrostunnel Recent extreme cold in Fairbanks got Ts down to -35C inside the Tunnel and check out this ice cracking! Had never seen this before. Likely contraction cracking due to extreme cold. Great example of ice wedge formation! R. Hessner pics
