Flew business class with @etihad recently – damaged my multiple bags – Asked for endless documents – Then refused responsibility Etihad sells “premium service” but delivers premium negligence. If this is how they treat business class passengers, think twice before booking.

Correct! Anthracyclines were originally discovered in Italy approximately 70 years ago by a company named Farmitalia. A member of the team went on expedition to Castel del Monte, in Puglia, in which soil he identified a strain of Streptomyces producing a substance with antitumoral proprieties: he called it “Daunomycin”, from the tribes of “Daunii” that used to live in Puglia during the Bronze Age. The first trials with daunomycin were conducted at the NCI of Milan and at MSKCC, describing the first clinical responses. Meanwhile, the team at Farmitalia kept working on the molecule, developing an even more potent analogue. They called it “Adriamycin”, from the Adriatic Sea that bathes the shores of Puglia, and that can be easily reached from Castel del Monte, where everything started. HER2-low: not only a breast cancer story. In this phase 2 trial enrolling patients with pretreated, HER2-low gastric/GEJ cancer, T-DXd (6.4 mg/kg) achieved an ORR of 26% in 2+ and 9.5% in 1+. Far from the DB-04 activity, but pretty intriguing nonetheless. ascopubs.org/doi/pdf/10.120…

@PTarantinoMD Great points! Underlying tumor biology likely potentiates the payload activity, irrespective of the target antigen level, which is probably what is being seen in patients. We proposed this for a different ADC - biorxiv.org/content/10.110… Having hard time to convince reviewers :)

@PingMu_PhD Nice to see growing evidences in support of important role of IFN/JAK/STAT signaling in prostate cancer. I presented some of our initial findings in 2017 at AACR (aacrjournals.org/cancerres/arti…). The textbook JAK/STAT pathway looks straightforward but is super-complex.
