@Tim_Lohoff @zhaoweiasu How does one define an endogenous target site? Do you still need to include a promoter into the payload with this system? Or can land under control of WT promoter?

@BowTiedBiotech @zhaoweiasu Seamless can program recombinases to almost any endogenous target site, without the need for a prior installed landing pad. The site-specific recombinases can ultimately catalyze inversions, insertions, excisions, and exchange reactions. Stay tuned!

@BowTiedBiotech @zhaoweiasu Just to clarify, Seamless is using recombinase-based gene editing, not retrotransposons, even more versatile 💪🏼

@zhaoweiasu I have many questions about retrotransposon specificity - it would seem to me they limited to landing within safe harbours which comes with risk...

Wow 🤯 Congratulations @felix_lansing and team 🎉 Amazing work. Looking forward to following the path of designer-recombinases towards the clinic 🩸💪🏼 Keep the exciting work up! 🍀 Check out the great tweetorial below 👇

Our work just got published in @NatureComms. We developed a dual designer-recombinase system that efficiently corrects a 140 kb inversion frequently found in patients with severe Hemophilia A. 1/11 @BuchholzLab_TUD @TakebeLab @tudresden_de @Medizin_TUD rdcu.be/cFmeD

Paper alert! 🎉 By combining SeqFISH and scRNA-seq data we've generated a spatially-resolved map of cell types and gene expression in the early mouse embryo. doi.org/10.1038/s41587…
