Overall, we showed that the mechanics of PCDH15 are modulated by Ca2+ and that PCDH15 has the appropriate stiffness to be part of the gating spring. The mechanics of the mutant suggest it is unable to convey adequate tension to the channels, likely the mechanism of deafness.
