🧬 If polyphenols slow aging, they probably aren't doing it by acting purely as antioxidants. They interact with multiple core aging pathways at once: mitochondrial function, inflammation, autophagy, proteostasis, epigenetic regulation, and cellular senescence. That matters because aging is a systems problem, not a single-pathway one. Not all p16-senescent cells are born equal. doi.org/10.1016/j.cell… In this elegant study targeted ablation of p16-positive macrophages or EC cells has the opposite effect on liver injury outcome. The elimination of p16+ Mф diminished fibrosis and the elimination of p16+ ECs augmented it. This study together with other works on the heterogeneity of senescent cells raises the question if non-selective senolysis is an appropriate approach to cure aging. (@LidskyPeter ) Senescent cells slowly turn out not to be pure evil, but like, everything else, partially adaptive and partially maladaptive context-specific respons to a stress. It also explains why senolytics fail to prolong mice's lifespan more than 15-20%.
