Long-Term Transcriptomic Reprogramming in Peripheral Blood Mononuclear Cells of Severe COVID-19 Survivors Reveals Pro-Oncogenic Signatures and Cancer-Associated Hub Genes 🔥The plot thickens……”severe COVID-19 infection may establish a systemic milieu favourable to cancer development or recurrence.”(Turkey) ➡️Authors provide the first evidence of a persistent pro-oncogenic transcriptomic signature in peripheral blood mononuclear cells (PBMCs) one-year post-severe COVID-19. ➡️RNA-seq revealed enriched cancer hallmarks: evading cell death, proliferative signaling, and invasion/metastasis, driven by hub genes like histone variants (H3F3C, HIST2H3D), transcription factors (JUN, EGR1), NOTCH1, and SQSTM1/P62, suggesting systemic epigenetic reprogramming primes tumorigenesis. ➡️Key Hub genes H3-5/H3F3C, H3C13/HIST2H3D, JUN, EGR1, NOTCH1, SQSTM1/P62 link inflammation, autophagy, and tumorigenesis. ➡️Systemic IL-6-driven awakening of dormant cancer cells, EMT promotion, NETs reactivation and epigenetic reprogramming via JUN/EGR1 and histone variants. ➡️Study aligns with studies showing SARS-CoV-2 accelerates metastasis (Chia et al., 2024) and creates inflammatory memory (Qian et al., 2025). ➡️They also have a small milder (non-pneumonia) C19 cohort in this study, that shows some persistent transcriptomic changes and cancer pathway enrichments, though less pronounced than in severe cases. ➡️So, COVID-19 primes a possible systemic tumor-promoting environment. ➡️"This study provides robust transcriptomic evidence that severe COVID-19 infection leaves a permanent, systemic, and pro-oncogenic immunological scar that persists long after infection and can increase cancer risk." ➡️"The identification of specific molecular targets like SQSTM1/P62 and the enrichment of cancer-related pathways elucidate the biology underlying this process." ➡️"Our findings, combined with the powerful evidence from both clinical observations and large human cohorts as well as experimental models, definitively underscore the critical importance of long-term oncological surveillance for patients who have recovered from severe COVID-19 for potential malignancy development or recurrence."
