Reason for $sava placebo out performance - Low pTau to MMSE scores Lower Death Rate on simufilam .3% vs. 1.1% in donanemab placebo - Interesting Outcome to Analyze. The RETHINK was only 12 months as compared to 18 months in the other study. I think the entire $sava simufilam placebo "out performance" for RETHINK phase 3 was based on the population having on average a lower pTau than a typical population for their MMSE scores. When the numbers are announced for the placebo pTau217 for Phase 3, I predict about 5.82 pG/mL. I think it was the screening that caused it to be lower in OL and in RETHINK. I think the outperformers in OL were based on uneducated Hispanic patients getting better at taking ADAS-Cog tests vs simufilam effectiveness in addition to the effects from low pTau217. Here is a chart of placebo donanemab with ADAS-Cog 13 1 year scores on the x and pTau217 baseline on the y. It has an R2 of .9999. Change in adas-cog13 is 100% correlated to pTau217 baseline scores. Has anyone seen other research that can prove this out? Shorts were right about the drug and it's likely match to placebo, but they never brought up the reason the performance was better than historical MMSE placebo scores. I am nearly certain the outperformance was driven by pTau217 and 181 being LOW relative to a typical MMSE in the population. I bet this is proven out in the deep analysis of the data by $sava. As for falls and other symptoms that families were reporting as improving, I do not know how to explain the one off cases, but the treatment and placebo of simufilam had the exact same Fall % leading to discontinuation. Death Rates for simufilam simufilam did have fewer deaths of .3% vs. .8% in placebo and 1.1% in donanemab. I would be interested in seeing this survival rate in the total data vs. the normal AD population. Donanemab had 1.9% of patients die on treatment and 1.1% die on placebo. Death rate by baseline pTau217 would be an interesting outcome to analyze
