Our results raise the attractive possibility of combining PI3Kα inhibition with the food supplement NAC, especially in patients with drug-resistant metastases associated with NF1 loss. This warrants further investigation in clinical trials. 10/10

We find NF1 more frequently mutated in metastases than in primary tumor samples. NF1 KO cells also show reduced sensitivity to ribociclib and fulvestrant. Co-treatment with NAC sensitizes cancer cells with NF1 loss to ribociclib and partially to fulvestrant. 9/10
