Nearly 20 years ago, researchers discovered something fascinating: some people are naturally protected from heart attacks because one of their PCSK9 genes is turned off, leading to low cholesterol from birth. That observation sparked a simple but ambitious question: can we give that same protection to others? Results from the Heart-2 Phase 1 trial released today show early clinical evidence that could open the door to new possibilities. These are results in 35 people and we have a lot left to learn. But, the results could potentially mean an entirely different way of treating high cholesterol — one infusion, lifelong cholesterol lowering. 62% LDL drop is basically rosuvastatin plus ezetimibe. The win is adherence, not potency. After twenty years of watching patients stop their pills, I get the appeal. But permanent liver editing demands long-term safety data.

Would you permanently edit the DNA of cells in your liver in order to lower your LDL cholesterol and your risk of a heart attack? Eli Lilly spent $1 billion a year ago to Verve Therapeutics, a company founded by cardiologist and geneticist @skathire on just that idea. This morning early data on the company's new experimental gene editor were published in the New England Journal of Medicine and presented at the annual meeting of the European Atherosclerosis Society in Athens. The treatment lowered LDL, which we know causes heart attacks, by 62% -- about the same amount as 10 mg of rosuvastatin plus ezetimibe. But Kathiresan points out to my colleague Jason Mast that constant LDL-lowering should work better than a pill patients often stop taking. “I treated patients for about 20 years, and you know, patients ask me all the time whether they’ll be on cholesterol medicine for the rest of their lives,” Kathiresan said. “And I think with VERVE-102 for the first time, I can tell them maybe not.” Cardiologists say it will take time, data, and experience to know if this approach is right for some people. There were no treatment-related serious adverse events in the Phase 1 study — a notable finding, given that Verve had to shelve its first candidate due to safety concerns.

Eli Lilly has done it. They've gone and made what seems to be a powerful, permanent gene therapy for LDL cholesterol. That means they'll be able to effectively prevent most heart disease with a single infusion!

The universe will make sure you'll win if you just keep going. not because life suddenly becomes easier overnight, but because the moment you truly believe in yourself, you stop moving like someone who’s waiting to fail. most people think success belongs to the smart, lucky or talented people. but actually, a huge part of it belongs to the people who refused to stop before the results showed up. every good thing in your life once existed as something uncertain, something invisible. there were moments where nothing made sense yet but you didn’t quit just because you couldn’t see immediate proof. you kept going anyway, and eventually you got to see it all with your own eyes. so keep showing up. keep visualizing. keep moving like the life you want is already making its way toward you. appreciate that you are constantly evolving and that this position you are in right now is not your final destination. cause the universe responds differently to people who trust themselves deeply. people who continue anyway. people who stay committed to the vision even when they have no evidence yet besides faith. what’s meant for you does not always arrive instantly. sometimes life is preparing you mentally before it delivers what you asked for. sometimes the delay is building the version of you capable enough of handling the blessing properly once it arrives. “every version of you that suffered was shaping the version strong enough to receive what you asked for.” so stop acting like your dreams are impossible just because they haven’t happened yet. keep believing. keep working. keep aligning yourself with the version of you that already has it. your future is responding to the energy you repeat every single day.

This is what I was saying earlier today. As we grow older, we think we should have be calmer or made better decisions. Pep came into Barcelona and immediately said he’s didn’t want Ronaldinho, Eto’o and Deco, he wasn’t ready to give them a chance. Same way he arrived at City and was clear he didn’t want Joe Hart…but as he grew as a manager, he would do some retrospection and wish he made better choices. Those who will benefit from his better version are the players at the next club he’s going. A typical example is Sir Alex and Beckham. Sir Alex learnt struggled at the end to manage such players, but when Cristiano came, he’d learnt. Don’t stop growing, evolve or die.
