The Weekly 🌱 New: Increased: $nktr, $bcrx Unchanged: $abvx, $cogt, $ewtx Decreased: Closed: $cytk I’ll start with $bcrx , I wrote about it earlier in the week and with $kalv taken out, leaves you with $bcrx with a strong upside case in HAE. I do think Navenibart has a very good case to becoming a BIC long acting injectable. Ph3 topline should be early next year and if data confirms the ~90% attack reduction profile, $bcrx would own the two most pt friendly modalities (oral + 3 to 6 month injectable) in HAE with commercial infrastructure already in place for the injectable launch. $nktr I got assigned the remainder of my 1000 contracts at 105 therefore an increase this week (without any fat candles). Resolution/progression of the $lly litigation remains on the agenda. Whilst Rezpeg remains a novel MOA with the most encouraging data of any Treg agent in inflammatory skin disease to date. Durability + quarterly dosing is a real differentiator. Someone mentioned the cancer risk on here and I would just like to say cancer risk is a theoretical concern for EVERY new I&I MOA. No such signal for rezpeg through >1000 patients and no known cancer syndrome from TReg/IL2 mutations in human epidemiology. Building on $abvx which everyone seems to be waiting for… Of the major UC maint trials (ACT, PURSUIT, GEMINI, ULTRA 2, TOUCHSTONE, OCTAVE) only OCTAVE (tofa) is a clean comp for obe. Tofa and obe are both oral small molecules. Tofa was ~50% advanced therapy exp, $abvx 47.3%. Similar steroid use. And the same maint design: induction responders randomized to stay on drug or switch to pbo (withdrawal). $abvx puts 678 responders into exactly that. When trying to do an apples for apples comp I realised $abvx scores on mMayo, tofa on full Mayo. But there is a published mMayo rescoring of OCTAVE. journals.sagepub.com/doi/10.1177/17… Level on mMayo. $abvx induction 50mg: 21.7% / 19.8% vs pbo 2.5% / 6.3% across the two studies. Tofa pooled induction on mMayo: tofa 10mg 24.8% vs pbo 7.7% (was 17.6% / 6.0% on full Mayo). Obe pbo adj induction remission is 16.4% (20.8 vs 4.4 pooled). Tofa is 17.1% (24.8 vs 7.7). But note that $abvx data is NOT like tofa and enrolled JAKi failures (it was the first JAK). $abvx built the largest JAKi failure cohort yet. If obe’s maint delta lands near tofa’s, it did so in a tougher pop. So we can use tofa ph3 as a guide to maint , wk52 on mMayo: pbo 12.1%, tofa 5mg 35.9%, tofa 10mg 42.1%. Same scoring, same design. Giving us ~12% as the empirical floor for a withdrawn responder pbo arm in an oral small mol. That is where maint pbo should land for $abvx. My base case for $abvx active 38% to 48%, pbo 12% to 15%, delta mid 20s to low 30s. Ph2b durability data leans toward the top of that range. Someone said to BP will focus on the absolute remission I think remission delta is more important here because absolute remission is suppressed by the mandatory steroid taper and the refractory mix. The delta is the clean efficacy signal and the regulatory and commercial yardstick. For instance a 40% / 13% readout and a 40% / 20% readout share a headline but are not the same drug. The underrated part I mentioned above also is breadth and durability. Obe works in bio exp and JAKi exp patients (where S1P modulators are far weaker), deepens over time (ample data supporting this) holds remission at a lower 25mg dose out to multiple years AND has arguably the cleanest safety of any oral UC mechanism. If the delta confirms at 25 to 30 pts with clean safety, that broad, oral, safe positioning is worth more than another 5 pts of remission. So anything over 25% delta this should rerate much higher Oh and I closed out $CYTK because I needed to put up margin due to being assigned $nktr shares Will revisit when liquidity returns.

Sure persistent inflation and higher oil could derail markets but nothing to worry about might be my largest worry. As long as cash is on the sidelines and shorts press bets it creates a a perfect structure for markets to continue north. This is why I’ve kept it simple, bought every dip and tried to not be too macro. If I was a macro trader I’d be homeless. One day the macro will over power the bull, no clue when. So as they say steady as she goes.

I'm losing lots and lots more followers today because I am glad Thomas "Look at ME!" Massie lost. So I might as well go all in and purge all the freaks from my follower base. There are hundreds of nations and thousands of interest groups that constantly seek to influence American public policy. If the only nation doing that which you are bothered by is Israel, and if the only PAC doing that which you are bothered by is AIPAC, and if you drone on incessantly, endlessly and vigorously about Israel/AIPAC and no OTHER NATION OR PAC, rational, intelligent people know why that is. It's because you are an antisemite. And if you want to say "Oh you are so wrong, I'm bothered by ALL nations and PACs attempting to influence US policy," but literally the only time you ever mention it in your X feed is in the context of Israel, you are an antisemite. I've been praying the past few weeks. Unsure why. There's good evidence behind prayer. It mimics breathwork, calming the nervous system, dropping cortisol, and quieting the brain. Daily prayers show lower depression, anxiety, and pain. I'd like to develop a prayer practice. Growing up, the protocol was written for me. Explaining whom to pray to, the structure of the prayer, and the boundary conditions. I don't really know how to pray now.
