Modern GWAS can identify 1000s of hits but it can be hard to turn this into biological insight. What key cellular functions link genetic variation to disease? I'm excited to present our new work combining associations + Perturb-seq to build interpretable causal graphs! A🧵

1/ We report @ImmunityCP that the PD-1:PD-L1 axis promotes the development of B-cell memory and antibody responses in mice and humans. To our surprise, the pro-B-cell effect of PD-1 and PD-L1 is, at least partially, mediated in a B-cell-intrinsic manner. cell.com/immunity/abstr…

Imagine a patient at initial presentation with an immune-mediated disease; rather than focusing on tissue/antibody-based diagnosis, you treat based on a test that captures the patient's global immune health. Another great step in that direction from my colleagues @tsanglab

1/12 Check out this paper that came out last week from our pan-monogenic project, which utilizes diverse natural single-gene defects in humans to study and assess the immune health of both patients with disease or those clinically healthy. nature.com/articles/s4159…

There's been amazing coverage of our new paper @Nature nature.com/articles/s4158… but I wanted to put together a walk-through (and make this accessible as possible). It's a bit long so get a cup of tea! The full version is open access so pls do have a read if you are interested...

It's so meaningful to share this chapter of work with my colleagues @TsangLab with my wife Sarah, who created the cover for @ImmunityCP. Days spent working together while in grad / art school, overlooking our garden in Cambridge, were some of the happiest in my life :) 1/

Immunology Twitter, I need your help. I am looking for a scRNAseq dataset for mouse ILCs (if gut specific even better). Can someone highlight some reference?

Using spatial metabolic flux we identified the conditions to preserve discarded human donor kidney’s for at least 4 days ex vivo. Such an approach also holds the potential for resuscitation and could increase the window for organ allocation. rdcu.be/dHL6W

Our paper @ImmunityCP reveals a human baseline setpoint state (ie, "temperature" under which immune responses are initiated) linked to better responses to flu vaccination resembles cell states induced early and uniquely by the AS03 adjuvant. 1/n
