Thrilled to see our work out in @naturemethods today! We applied prime editing to multiplexed dropout screens and benchmarked the tool’s ability to induce phenotypes when installing premature stop codons in essential genes.

But wait! What about the host sequences? We describe a total of 8 different workflows for the masking of host sequences. We found that many viruses, including known infections, will have some reads that also align to the host, making this a complicated issue.

We expanded kallisto to translated alignment (nucleotides <-> amino acids), and used it to detect novel viruses in RNA seq data. The single-cell resolution allowed us to determine whether the presence of these viruses affected host gene expression. 🧵 biorxiv.org/content/10.110…
