Update: celebrated too early. Had to use all the DNA I got per line and both sets of genotyping PCRs are inconclusive for different reasons 🙃 science is fun sometimes
When you make an accidental mistake in prepping gDNA, continue anyways, and despite very low concentrations the nanodrop still gives you that sweet 160 curve (these are transfections I can’t easily get more cells 😬)
@JPeterMcgee Definitely not the worst mistake. Just today I threw my western membrane in the trash and lovingly put my leftover milk on the rotator in the fridge. (Thankfully the falcon tube was shut and the western turned out ok😅)
Hey kids just a reminder that everyone can make simple mistakes now and again, like plating two different transformations on the same plate 😬 (I “rinsed” both tubes with LB Broth and plated them separately with plenty of colonies on each plate so not the end of the world ~yet~)

I think a major shortcoming stems from the tendency to measure what is easiest rather than what is biologically most relevant.

@MelodyWYY@Melterialsgal If you can, try and talk to people actually in the lab, as many as possible. It's hard for multiple people to hide serious problems in the environment. The other piece of advice is to take your time. Don't jump to join a lab because you might lose your shot, explore your options.
