Milwaukee Road SD10 #561 is tied down for the weekend at Mineral Point, Wisconsin on Sunday, July 9, 1978. This town sat at the end of the Mineral Point Branch and was heavily weed-choked, as noted by the downed foliage on the locomotive's walkways. What's now for the Amyloid Hypothesis in #Alzheimers? Several recent studies using monoclonal antibodies show evidence that removing aggregated Aβ from the brains of symptomatic patients can slow the disease progression. Yes, despite the clinical benefit of these trials still being modest, disease-modifying treatments for Alzheimer's are feasible, and they are going to happen more consistently within the next few years. I and others are focusing on understanding how soluble amyloid species, not senile plaque or large fibrils, can play a crucial role in synapse loss and neurodegeneration in Alzheimer's. Oligomeric amyloid, for example (🔵 in the image below), triggers extensive glial activation (🟠astrocytes) in the human brain. By using promising antibodies and optimizing the timing for therapeutic intervention, we might be finally close to helping patients with Alzheimer's and similar neurodegenerative diseases.
