What factors should clinicians consider when deciding to use a drug off-label? Yesterday I sat down with master repurposing clinician Dr. Luke Chen to discuss this. Here are several factors that come to mind: 1. Safety Profile: How safe is the drug? The patient’s overall health and potential risks are crucial considerations. 2. Severity of the Disease: How severe is the disease? The urgency of the situation often influences the willingness to try new treatments. 3. Mechanism of the Drug: Is there a plausible connection between the drug’s mechanism and the disease? Understanding this connection is crucial. 4. Clinical Data: Is there clinical data suggesting the drug has been effective in similar cases? Prior usage can provide valuable insights. 5. Cost of the Drug: The drug’s cost can be a significant factor. If it's exorbitantly expensive, it might change the decision-making process. 6. Patient’s Current Condition: Consideration of the patient’s current state in their disease course is essential. The decision might differ based on their immediate health status. My passion for finding new uses for existing drugs started long before #chasingmycure While my mom was battling brain cancer, her doctors gave her a high dose of corticosteroids which pulled her out of a coma and allowed us to have 2 additional months with her before she passed away. Those two months were some of the most special two months of my life and it was all thanks to a drug that wasn't made for her disease. Fast forward years later, I nearly died five times from a rare and deadly illness before discovering, Sirolimus, a drug that wasn’t intended for my disease, giving me 9.5 years that I never expected to have. These personal encounters highlight why I am on a mission to find new uses from existing drugs. Alongside my teams at @everycure, @pennmedicine, and @curecastleman nothing can stop us from discovering cures for patients who need them most. #MedTwitter

Funds raised through Light The Night allow LLS to fund treatments for patients who are suffering from all forms of 🩸 cancers. Since 2017, LLS has helped advance 85% of the blood cancer treatment options approved by the FDA.

@DavidFajgenbaum @ElevatePrize Thank you for bringing attention to rare diseases! The fact is that many if not most rare diseases are not listed as an approved indication under any therapeutic, therefore coverage of therapy usually denied even when evidence supports the therapy.
