#ARTNetPublication New research in @CR_AACR reveals a promising strategy to overcome radioresistance in lung cancer. @BoyiGan, @ACKoongMDPhD, @StevenLin_MDPhD, @Dadi_Jiang, and colleagues identified DHODH as a key metabolic driver that protects tumor cells from ferroptosis while supporting DNA repair after radiation. Combining DHODH inhibition with radiotherapy and IFNγ/anti-PD-1 immunotherapy dramatically improved treatment response in preclinical models, highlighting a promising therapeutic strategy for lung cancer and potentially other solid tumors. Congratulations to @BoyiGan, @xingxing_zang, @StevenLin_MDPhD, and all co-authors on this outstanding study published in @CellCellPress! The team uncovers a powerful two-way connection between antitumor immunity and cuproptosis: CD8+ T cells enhance tumor sensitivity to copper-dependent cell death through the IFNγ–STAT1–IRF1–FDX1 axis, while cuproptotic tumor cells stimulate immune responses by acting as immunogenic cell death. Importantly, combining a cuproptosis inducer with anti-PD-L1 therapy helped overcome immunotherapy resistance across multiple preclinical models.
